Metformin induces autophagy of cisplatin-resistant human gastric cancer cells in addition to apoptosis

چکیده

Metformin has been used to treat cases of type 2 diabetes mellitus, and mounting studies have shown that metformin can act alone or in synergy with other anticancer agents achieve anti-cancer efficacies on various types tumors. However, the role either inducing autophagy cisplatin-resistance human gastric cancer (GC) cells never examined. The study established a cisplatin-resistant GC cell line investigated effects it. results demonstrated treatment concentration-dependently suppress viability confluence cells, while having no primary stomach epithelial (HPSEC). For first time, we found significantly increase acidic vesicular organelles (AVO) level decrease acridine orange (AO) spontaneously cells. Thus, further checked markers, Atg5, Atg12 LC3-II, which showed indeed induced In addition, 3-Methyladenine (3-MA) rescue metformin-induced autophagy. At same induce alterations apoptosis-associated signal molecules, such as caspase-3 caspase-7 activities. Overall, pilot provided evidence for addition apoptosis, making it an effective drug therapy GC. Killing non-toxic via both apoptosis might extend its usefulness our fighting chemo-resistance

Liquiritin induces apoptosis and autophagy in cisplatin (DDP)-resistant gastric cancer cells in vitro and xenograft nude mice in vivo

Gastric cancer is reported as one of the leading factors resulting in tumor-related death worldwide. However, the therapies to suppress gastric cancer are still limited and the emergence of drug resistance makes it necessary to develop new and effective anticancer drugs and combinational chemotherapy schemes. Liquiritin (LIQ) is a major constituent of Glycyrrhiza Radix, exhibiting various pharm...

MicroRNA-101 induces apoptosis in cisplatin-resistant gastric cancer cells by targeting VEGF-C.

Deregulation of microRNAs (miRNAs) is known to be associated with drug resistance in human cancers. However, the precise role of miR‑101 in the cisplatin (DPP) resistance of human gastric cancer cells has not been elucidated, yet. The present study revealed that miR‑101 was markedly downregulated in gastric cancer cell lines compared to that in the normal gastric mucosa epithelial cell line GES...

evaluation of antagonistic effects of metformin with cisplatin in gastric cancer cells.

metformin has recently been introduced as an anti-cancer agent. in this study, we evaluated the effect of metformin and metformin/cisplatin on human gastric mkn-45 cell line. when we used metformin alone, it could inhibit proliferation and induce apoptosis, but it diminish anti-proliferative effects of cisplatin when they are used in combination. further, we checked mrna levels of survivin, mto...

Cryptotanshinone induces ROS-mediated apoptosis in human gastric cancer cells

Cryptotanshinone (CT), isolated from the plant Salvia miltiorrhiza Bunge, has been reported to have potential anticancer effects on human prostate and breast cancer cells. However, the mechanisms of action of CT on gastric cancer (GC) cells are not well understood. Here we investigated the antitumor effects of CT on GC cells and its possible molecular mechanism. We found CT suppressed viability...

Chestnut extract induces apoptosis in AGS human gastric cancer cells

In Korea, chestnut production is increasing each year, but consumption is far below production. We investigated the effect of chestnut extracts on antioxidant activity and anticancer effects. Ethanol extracts of raw chestnut (RCE) or chestnut powder (CPE) had dose-dependent superoxide scavenging activity. Viable numbers of MDA-MD-231 human breast cancer cells, DU145 human prostate cancer cells,...
